The condensation enzyme CcbD catalyzes an amide bond formation between the carrier protein-tethered proline and thiooctose in the biosynthesis of lincosamide antibiotics. Here, we report the structure-function analyses of CcbD by in vitro characterization, crystallization, and mutagenesis.
Structure-function analysis of a new type of endoperoxidase
Endoperoxidase NvfI catalyzes the incorporation of “three” oxygen atoms, including an endoperoxide and a hydroxyl group, into its substrate asnovolin A, in a single enzyme turnover reaction through a different mechanism from those of previously characterized endoperoxidases.
